Cooper, Robert C. (Michigan State University, East Lansing). Evidence for the presence of certain tricarboxylic acid cycle enzymes in Thiobacillus thioparus. J. Bacteriol. 88:624-629. 1964.-Various tricarboxylic acid cycle enzymes appear to be present in Thiobacillus thioparus. Cell-free extracts of T. thioparus were active for a number of tricarboxylic acid cycle enzymes, including aconitase, isocitric dehydrogenase, and malic dehydrogenase. Tests for the presence of fumarase and the condensing enzyme, citrogenase, were inconclusive. Citrate was shown to be active in the metabolism of T. thioparus, but the actual mechanism involved in its formation was not clear. The enzyme, isocitratase, appeared to be absent. Evidence for the presence of succinic dehydrogenase was found in experiments with whole cells. From these results, it would appear that T. thioparus has a terminal respiration pathway similar to that found in many heterotrophic microorganisms.

The oxidation of thiosulfate and phosphorylation in extracts of Thiobacillus thioparus.

Thiobmillus thioparus is an autotrophic microorganism that obtains the energy required for growth by the oxidation of reduced inorganic sulfur compounds to sulfate. When it is grown on high concentrations of thiosulfate, the organism accumulates elemental sulfur and produces sulfate. The pathway of thiosulfate oxidation has been essentially unknown; however, it has been postulated that tetrathi...

Assimilation and metabolism of exogenous organic compounds by the strict autotrophs Thiobacillus thioparus and Thiobacillus neapolitanus.

The assimilation and utilization of the individual carbon atoms of pyruvate and acetate by cells of Thiobacillus thioparus and T. neapolitanus, in the presence and absence of an energy source, were studied by use of radioactive substrates. Both organisms produced (14)CO(2) from (14)C-labeled pyruvate, but more came from carbon 1 than from carbons 2 or 3. The conversion of the carbons of acetate...

Permanent draft genome of Thiobacillus thioparus DSM 505T, an obligately chemolithoautotrophic member of the Betaproteobacteria

Thiobacillus thioparus DSM 505T is one of first two isolated strains of inorganic sulfur-oxidising Bacteria. The original strain of T. thioparus was lost almost 100 years ago and the working type strain is Culture CT (=DSM 505T = ATCC 8158T) isolated by Starkey in 1934 from agricultural soil at Rutgers University, New Jersey, USA. It is an obligate chemolithoautotroph that conserves energy from...

The oxidation of tricarboxylic acid cycle compounds by Penicillium chrysogenum.

The presence of a tricarboxylic acid cycle in Penicillium chry8ogenum, as yet, has not been demonstrated conclusively. Ajl (1951) stated that of the mechanisms proposed for oxidation of the 2 carbon fragment in mold metabolism, the evidence for a tricarboxylic acid cycle is the least convincing. This paper presents a study of some of the enzymes of the tricarboxylic acid cycle in cell-free prep...

Reversible transdominant inhibition of a metabolic pathway. In vivo evidence of interaction between two sequential tricarboxylic acid cycle enzymes in yeast.

The enzymes of the Krebs tricarboxylic acid cycle in mitochondria are proposed to form a supramolecular complex, in which there is channeling of intermediates between enzyme active sites. While interactions have been demonstrated in vitro between most of the sequential tricarboxylic acid cycle enzymes, no direct evidence has been obtained in vivo for such interactions. We have isolated, in the ...
